摘要:
利用RT-PCR技术首次克隆获得了乌鳢(Channa argus)的β-肌动蛋白基因cDNA序列.结果显示,乌鳢β-肌动蛋白的cDNA序列全长1 813 bp,包含一个1 125 bp的开放阅读框(ORF),共编码375个氨基酸.蛋白序列分析显示,乌鳢β-肌动蛋白的相对分子质量为41.731 kDa,等电点PI为5.16,由20种氨基酸组成(甘氨酸数目占比最高为7.73%,色氨酸数目占比最低为1.07%),含有3个actin信号位点.蛋白同源性分析显示,乌鳢β-肌动蛋白与大西洋鲑、虹鳟、莫桑比克罗非鱼的同
Abstract:
A 1813 bp full-length cDNA sequence of β-actin gene was obtained from Channa argus using RT-PCR technique, which contain a 1 125 bp open reading frame(ORF)encoded a 375-amino acid residue. The predicted protein molecular weight was 41.73 kDa, and the supposed isoelectric point(PI)was 5.16, which was composed of 20 kinds of amino acid and included 3 actin signal locus. Comparative analysis showed a high homology(more than 98%)of β-actin between Channa argus and other vertebrates, indicated that β-actin was highly conserved among different species. Phylogenetic analysis was performed on the basts of β-actin nucleotide sequence. The results showed that Perciformes and Salmoniformes clade were rooted with Channa argus, which indicated that the β-actin gene has a lower evolution rate in Channa argus than other Perciform fishes. Relative β-actin mRNA expression was detected in 11 tissues, results showed that the β-actin gene was widely and stablely expressed in all tissues, which indicated that it could be used as a candidate reference gene in other research.
